Played around some, and neither Replaygain, the Pre-Amp, or the EQ is working. To fix the issue, I tried changing my sound settings from pulseaudio to ALSA, disabling the volume control, completely uninstalling Strawberry (removing config files from .config as well). I can share the files I was using to test Replaygain if needed.

Strawberry - Version 0.9.2 (from the ubuntu PPA)
      OS: Pop!_OS 20.10

        All those features work here.
With the replay gain setting does not take affect before you stop/start playback.
The equalizer is possible to change while playing as long as you check the enable button before you start playback.

@jonas I realize that I didn't poke around with it enough, eq is working fine for me. When I was playing before too, i only tried turning the gain up, not down. I can't notice the effects when turning it up, but I can when turning it down?
Here's a video of me demonstrating the behaviour.
